We have investigated the surface plasmon enhancement of the GeSn p-i-n photodiode using gold metal nanostructures. We have conducted numerical simulation of the plasmonic structure of 2D nano-hole array to tune the surface plasmon resonance into the absorption range of the GeSn active layer. Such a structure is fabricated using FIB on a previously made photodiode for feasibility demonstration. The result indicates that the photocurrent of the diode can indeed be enhanced with the plasmonic...

An analytical study was made of the feasibility of using an MHD- augmented arc as the primary component of a large reentry vehicle test facility. Computer studies of the performance of MHD accelerator channels were made for systems having channel input power of 50 MW to 1,700 MW. It was determined that complete simulation of reentry conditions is not possible in a single extended- flow facility. The studies showed, however, that very large increases in performance over existing arc facilities...

Obesity, which induces low-grade inflammation, is a known risk factor for worse prognosis in many cancers including breast. We found that sphingosine-1-phosphate (S1P) produced by sphingosine kinases (SphKs) plays a critical role in obesity-related inflammation and breast cancer. Obesity increased S1P in the tumor microenvironment, as well as in the primary tumors. FTY720, a functional antagonist of S1PR1, dramatically decreased cancer progression by reducing expressions of SphK1 and S1PR1, and...

In this paper rank estimates, called WLS rank estimates and computed using iteratively reweighted least squares, are studied. They do not require the estimation of auxilliary scale or slope parameters nor do they require numerical search techniques to minimize a convex surface. The price is a small asymptotic efficiency loss. In the location model, beginning with a resistant starting value such as the median, the WLS rank estimates have good robustness and computational properties. The WLS rank...

To ensure sufficient capacity to handle unexpected demands for electric power, decision makers often over-estimate expeditionary power requirements. Therefore, we often use limited resources inefficiently by purchasing more generators and investing in more renewable energy sources than needed to run power systems on the battlefield. Improvement of the efficiency of expeditionary power units requires better managing of load requirements on the power grids and, where possible, shifting those...

Multi-lingual feature extraction using bottleneck layers in deep neural networks (BN-DNNs) has been proven to be an effective technique for low resource speech recognition and more recently for language recognition. In this work we investigate the impact on language recognition performance of the multi-lingual BN-DNN architecture and training configurations for the NIST 2011 and 2015 language recognition evaluations (LRE11 and LRE15). The best performing multi-lingual BN-DNN configuration...

This project is focused on Systemic Sclerosis (SSc), a progressive fibrotic disease characterized by skin fibrosis and damage to internal organs. While a wide range of environmental and biological triggers have been proposed, no definitive etiologic agents have yet been identified. Metagenomic analysis of non-human sequences in SSc RNA-seq data was used to detect microbial sequences in human tissues in an unbiased, quantitative manner. Our studies suggest that disease pathogenesis includes a...

Biosensors based on organic thin film transistor technology offer the exciting prospect of detecting and identifying peptides; biomarkers found in bodily fluids. This development would assist in the monitoring or identification of medical diseases and facilitate rapid, point-of-care testing. In particular, neuropeptide Y is a highly abundant neurotransmitter with links to many important physiological functions, including stress response. Its rapid detection provides an opportunity to identify...

In Year 2 of this award we examined whether duraplasty after acute thoracic spinal cord injury (SCI) in a porcine model could improve long-term functional outcome after injury. We finalized the 12-week PTIBS analysis as well as the somatosensory-evoked potential, which was recorded at baseline, 3 hours post injury and at the final week of the study (12 weeks post SCI). Our data suggest that duraplasty surgery might improve functional recovery early after SCI compared to SCI-animals without...

The majority time this reporting period was spent developing and testing the protocol of our study. This included training research staff, piloting study tasks, validating the quality of data collected. After developing our initial study tasks, we successfully enrolled and completed 36 participants over the course of six weeks, a total of twelve triads. These triads provided us with important information regarding the updating and fine-tuning of our protocol as well as our first steps for data...

The goals of this translational study are to create an animal model of joint injury and evaluate the impact of Vitamin D in prevention and progression of PTOA. Concurrently, we will evaluate a clinical cohort of USMA cadets treated for ACL tear, with pre- and post-injury serum 25-hydroxy-Vitamin D levels and correlation with joint space narrowing and biomarkers of cartilage injury. If Vitamin D supplementation can prevent the onset of often rapid joint destruction that occurs with PTOA, this...

The objective of this project is to discover and characterize molecules that inhibit breast cancer cell proliferation by maintaining activity of the retinoblastoma protein (Rb). Rb is inactivated to drive proliferation in normal and cancer cells by phosphorylation, which dissociates the E2F transcription factor from Rb. Our goal is to find and characterize molecules that stabilize the complex between phosphorylated Rb and E2F.

We devoted effort to both aims, to optimize methods and publish the first phase of efforts. SA1 is to understand the intrinsic mechanisms of dissemination by early-progressed cancer cells and how the microenvironment in these primary sites named P-TMEM (Primary Tumor Microenvironment of Metastases) contribute to early dissemination. We now have new data suggesting that a signature of early dissemination markers consisting on HER2HI/p-ATF2LO/E-cadhLO could identify motile early-progressed tumor...

Injury severity in blast induced Traumatic Brain Injury (bTBI) increases with blast overpressure (BOP) and impulse in dose-dependent manner. Pure primary blast waves were simulated in compressed gas shock-tubes in discrete increments. We evaluated 24 hour survival of rats in 0-450 kPa (0-800 Pas impulse) range at 10 discrete levels (60, 100, 130, 160, 190, 230, 250, 290, 350 and 420 kPa) and determined the mortality rate as a non-linear function of BOP. Using logistic regression model,...

A history of TBI increases the odds of developing AD by 2.5 times in the general population, and 4-6 times for military veterans. Although significant associations between mTBI and risk of AD have been observed, the precise mechanism by which TBI might lead to AD and/or AD-related symptoms are not yet understood. Histologically, AD is characterized by amyloid- and neurofibrillary protein aggregates, suggesting a loss of protein processing is a key feature of AD. MiRNAs are small non-coding RNA...

Traumatic eye injury (TEI) is one of the leading causes of monocular blindness in military personnel and young males worldwide. This profound and frequently irreversible posttraumatic loss of vision has a poor prognosis due to retinal cell death, scar formation, and lack of functional regeneration. Proliferative vitreoretinopathy (PVR), a form of intraocular fibrosis, is often the primary reason for the loss of vision after ocular trauma, and frequently occurs after blunt trauma and open globe...

DNA vaccine technology holds great promise as a platform for developing vaccines against both emerging and established global pathogens. Despite this potential, significant challenges impede the capacity of DNA vaccines to prevent disease in humans. Foremost amongst these, is the gap between remarkable results obtained in pre-clinical mouse models and relatively modest immunogenicity observed in humans. The present work is testing the hypothesis that skeletal muscle myocytes sense cytosolic DNA...

The purpose of this study is to investigate the role of parathyroid hormone-related protein (PTHrP) in cancer stem cell-driven triple-negative breast cancer. Our goal is to confirm PTHrP as a targetable molecular driver in order to develop better therapies against this notoriously hard-to-treat family of cancers. We have now constructed animal models with mammary epithelium fluorescent labelled cells to allow tracing of cancer stem cells from the primary mammary tumor to the metastatic site. We...

The study during the reporting period has used blood samples from leukemia patients who received therapeutic radiation for discovery of biomarkers. This could be used for exposure estimation in soldiers and emergency responders involved in nuclear war. The method for discovery and validation of biomarkers was optimized using samples collected from patients enrolled in two clinical trials. The utility of selected biomarkers identified from our earlier animal models has been validated and...

The medial olivocochlear reflex (MOCR), a feedback mechanism that controls gain of the outer hair cells, is thought to provide protection and enhancement for a listener in background noise. This listening advantage can be attributed to a suppression of auditory nerve firing, which results in an increased dynamic range for encoding a signal of interest (e.g. a tone or speech). The computational model presented here updates the work of Smalt et al. (2014), which added some of the dynamic...
