AB A nonconventional liposome compn. consisting of nonphospholipid lipids,
esp. cholesterol and cholesterol ester salts, are used for encapsulation
of drugs. They are useful for sustained release of steroids, and are
suitable for treatment of inflammatory, arthritic, rheumatoid diseases,
etc, esp. as aerosols for interstitial lung disease. Beclomethasone
dipropionate (I) 10 was incorporated into liposomes prepd. with Na
cholesterol sulfate 50 and cholesterol 40 mol %. Sustained release of I
was obsd. in rats following intratracheal administration, in contrast to
liposomes formulated with phosphatidylcholine and cholesterol.

AB The title spherules comprise nonionic or ionic amphiphilic lipids and
.ltoreq.50% cholesterol sulfate. A drug dispersion was prepd. by
homogenizing at 90. degree, a nonionic amphophilic lipid 7.6, Na
cholesterol sulfate 0.4, and . beta . -methasone 17-valerate 0.08 g with 20 g
H20, followed by the addn. of Me p-hydroxybenzoate 0.3, glycerol 5.0, and
H20 52.02 g. Subsequent homogenization yielded 0.2 .mu.m spherules, to
which perfume 0.4, Carbopol 0.4, triethanolamine 0.4 and H20 13. 8g were
added. The amphophilic lipid was R0CH2CHR10 [CH2CH (CH20H) O] nH (R =

dodecyl, Rl = equimol . mixt . of tetradecyl and hexadecyl, n = 5.5).

AB Liposome formulations comprising entrapped active ingredients, such as
amino sugars, and uses thereof are disclosed. In one embodiment,
intra-articular injectable formulations are provided. DSPC (33 .mu.M) and
cholesterol (33 .mu.M) were dissolved in CHC13-MeOH and the
solvent was evapd. N-Acetylgalactosamine in 0.85% NaCl soln. and mixed
with the above lipid to form the liposomes.

AB A compn. for controlling bacterial growth/colonization is provided. The

compn. comprises an enzyme, an anchor mol . coupled to the enzyme to form
an enzyme-anchor complex, with the anchor being capable of attaching to a
substrate proximal to a bacterial colony. The compn. is produced using
recombinant DNA techniques. In one form, the compn, is a fusion protein
coded for by a polynucleotide segment. The attachment to the substrate
permits prolonged retention time of the enzyme-anchor complex where the
bacterial colony is present to increase the effectiveness of the complex.
The invention is also for a method of controlling colonization of
bacterial plaque in the oral cavity, as well as a method of forming a
compn. for controlling the proliferation of bacterial colonies in the oral
cavity.

Purified wheat germ agglutinin (WGA) was derivatized with palmitic acid at
an av. stoichiometry of 1 fatty acid/dimer. Palmitoyl WGA was readily
incorporated into liposomes with a Na cholate dialysis method. The
acylation reaction was done in the presence of N-acetylglucosamine to
protect the active site of wheat germ agglutinin, and Na cholate (instead
of Na deoxycholate) was used throughout the procedure. Liposome bound WGA

caused agglutination of red blood cells at a concn. 8-fold lower than that
of the native lectin. Furthermore, enhanced binding of liposome -bound WGA
to mouse spleen cells was also obsd. Potential applications of the
liposome -bound lectin are discussed.

An effective process of facilitating the healing of wounds of the body
surface is described which materially reduces the time during which the
possibility of infection is greatest. In an example, 2000 g.
N-acetylglucosamine is added to 20 1. of 0.1% aq. saline soln. The
resulting soln. is filtered through a Seitz filter and stored in infusion
bottles each contg. 1 1 . of 10% soln. The soln. is useful for intravenous
administration to a human host having a surface wound. In addn. to a
remarkable improvement in healing time, such patients generally show a
sense of well being, maintain body wt . , and exhibit pos . N balance in
place of the usually expected neg. balance during the immediate
postoperative period; there is also a marked lowering of blood
cholesterol •

AB A novel group of compds . is disclosed for decorating the surface of
synthetic polymeric or tissue derived prostheses to prevent adverse
rejection events. The decorating mols. are obtained as derivs . of
naturally occurring polysaccharides, derivatized to provide functionally
reactive groups at the termini thereof, and the reacting with nucleophilic
or other groups on the surface of the prosthesis in a simple one step
reaction. Some of these reagents are useful in noncovalent adsorption to
polyolefinic or perf luorocarbon based materials. Finally, phospholipids

partially substituted with the nonantigenic polysaccharides provide a
superior bipolar component for liposome formation. Chondroitin
sulfate-modif ied-distearoyl phosphatidylethanolamine (I) was
prepd. by reaction of chondroitin sulf ate-CO-N-oxysuccinimide
with lyso-distearoyl phosphatidylethanolamine. Liposomes were prepd.
micro-f luidization (emulsif ication) of a compn. of l/hydrogenated soy
phosphatidyl choline/cholesterol in molar proportions of 5:55:40,
resp. It is anticipated that the blood circulation half -lives of the
biocompatible liposomes will be significantly longer than those of
liposomes formulated without the I.
